Im legendären Orient-Express passiert nachts ein spektakulärer Mord. Zu den Reisenden gehört auch der berühmte Privatdetektiv Hercule Poirot. Während der Luxuszug durch Schneeverwehungen in Jugoslawien festgehalten wird, kann er mit seinem scharfen Verstand ein außergewöhnliches Verbrechen aufklären. Istanbul, im Jahre 1935. Nach einem glänzend erledigten Auftrag in Jordanien will Hercule Poirot (Albert Finney) die Heimreise antreten. Für Signor Bianchi (Martin Balsam), Direktor der Eisenbahnlinie, ist es Ehrensache, dem berühmten Detektiv noch einen Platz im voll besetzten Erster-Klasse-Schlafwagen des Orient-Express zu beschaffen. Es wird eine unruhige Nacht für Monsieur Poirot. Einmal wird er durch einen unterdrückten Schrei aus dem Schlaf gerissen, dann wieder hört er, wie sich die exzentrische Mrs. Hubbard (Lauren Bacall) draußen im Gang darüber beschwert, dass ein Mann sich in ihrem Abteil versteckt habe. Am nächsten Morgen findet man den Amerikaner Ratchett (Richard Widmark) im Abteil nebenan ermordet auf. Er reiste mit seinem Sekretär McQueen (Anthony Perkins) und dem Diener Beddoes (John Gielgud) und hatte offensichtlich Grund, um sein Leben zu fürchten. Der unbekannte Mörder muss noch im Zug sein. Direktor Bianchi bittet Poirot, sich der Sache anzunehmen, während der Express in einer großen Schneeverwehung festliegt. Und der scharfsinnige kleine Detektiv merkt bei seinen Verhören bald, dass auch so illustre Mitreisende wie Prinzessin Dragomiroff (Wendy Hiller) und Colonel Arbuthnot (Sean Connery) oder die fromme Greta Olson (Ingrid Bergman) keineswegs über jeden Verdacht erhaben sind.
