Kurz nach dem Tod seines Vaters lernt der junge Schriftsteller Sal Paradise in New York den gleichaltrigen, lebenshungrigen Dean Moriarty kennen. Kurzerhand reißt Dean Sal aus seiner Trauer und der stockenden Arbeit an seinem Roman und nimmt ihn mit auf einen Roadtrip Richtung Westen. Berauscht von der unendlichen Weite der Landschaft, den Drogen, dem Sex, dem Jazz und ihren wilden Gedanken gelingt es Sal, seine Sorgen zu vergessen. Auf ihrer Reise stoßen sie auf neue Wegbegleiter und kehren ein bei alten Bekannten: Darunter sind Deans große Lieben Marylou und Camille, der unverwechselbare Old Bull Lee und seine Frau Jane sowie der gebildete und nachdenkliche Carlo Marx.
